Biological Description

ASIS-P020, published as Compound A2B57 (Tatum, Sawada et al. 2014), is a sirtuin inhibitor with high affinity and specificity for SIRT2 (IC50 of 6.3 µM for SIRT2, >100 µM for related SIRT proteins). Sirtuins, known as NAD+ dependent protein deacetylases, have been implicated in Parkinson’s Disease (Anderson and Maes 2014). SIRT2 has been shown to destabilize microtubules through deacetylation of ?-tubulin and influences gluconeogenesis by acting upon phosphoenolpyruvate carboxykinase (PEPCK)(Nakagawa and Guarente 2014). Novel SIRT2 inhibitors may function not only as tools for elucidating deacetylase effects but also as potential therapeutic agents. One such inhibitor, AGK2, shows a similar affinity for SIRT2, but less specificity with respect to SIRT1 and SIRT3. ASIS-P020 is a potent, specific inhibitor for SIRT2.
